RAPD analysis of interspecific hybrid between Gossypium arboreum and Gossypium stocksii

Abstract

An interspecific hybrid between G. arboreum x G. stocksii was obtained by pollinating previous day bagged flowers of G. arboreum Genetic Male Sterile line MPKV GMS with pollens from wild Gossypium stocksii. The hybridity was confirmed by Random Amplified Polymorphic DNA (RAPD) analysis in the hybrid plants in comparison with their parents. For RAPD analysis, seven random primers were selected and seven types of markers identified of which Type IV markers were used in determining the hybrid nature of the offspring.8.8% of Type IV markers present in the offspring were shared with the male parent confirming the hybrid status by ruling out the possibility of self fertilization and pollination.
