Abstract

A specific, sensitive and reliable radioligand assay for plasma dehydroepiandrosterone (DHA) and its sulfate has been developed. Antisera were obtained by immunizing rabbits with a DHA-17-albumin conjugate. DHA was separated from cross-reacting Δ5-steroids by thin layer chromatography. DHA-sulfate was solvolyzed prior to chromatography. Separation of antibody bound and free steroid was achieved with γ-globulin-dextran-coated charcoal. The standard curve was linear on a logit-log plot from 0.1 to 10 ng.
