Abstract

Ultrasonically induced hyperthermia (4.75 MHz, 43°C-47°C) and proton irradiation (160 MeV, 2000 rad-5000 rad) were evaluated alone and combined to tread experimental choroidal Greene melanoma in rabbit eyes. Threshold tumoricidal doses were established for each modality. As known from proton irradiation in human choroidal melanoma focused ultrasound was found to deliver well-localized energy to an intraocular tumor. Therapy was performed combining subthreshold does of heat (43°C) and radiation (1000 rad-3000 rad). Complete tumor control was obtained in the majority of all eyes treated. These results indicate that hyperthermia potentiated for tumoricidal effects of radiation, while sparing normal ocular structures. Therefore, we believe that experimental hyperthermia is suitable as an adjuvant treatment modality. This shows that ultrasound hyperthermia has the potential to increase the efficacy of proton irradiation by lowering radiation doses and thus decreasing posttreatment ocular morbidity in human intraocular malignancies.
