In an attempt to demonstrate an effect of a decrease in portal venous osmolarity in promoting an enhanced water diuresis in man, 10 volunteers were given 500 ml loads of .45% NaCl orally and intravenously. Paired observations at 20-minute intervals showed significant increase in free water clearance following oral loads at 80 minutes only (p<.05). Urine volume between the 2 routes of administration showed no significant difference. Doubling the size of the load or changing the order of administration did not enhance the response to oral loading. No conclusive evidence for existence of a hepatic osmoreceptor in man was obtained.
