Abstract

Glucocorticoid receptors of polymorphonuclear leukocytes (PMN) have been characterized and compared with those of mononuclear leukocytes (MNL). Cytosol receptors from PMN and MNL have similar binding affinities for [3H]-dexamethasone (Dex). nearly superimposable sedimentation profiles on sucrose density gradients, and similar elution patterns from DEAE-Sephadex. Thus, the glucocorticoid receptors of MNL and PMN have similar biochemical properties. We have developed a simple method for preparing leukocytes from 20 ml of blood to characterize and quantify the glucocorticoid receptor. The Scatchard analysis of [3H]-Dex binding to intact leukocytes shows a single class of binding sites; the apparent Kd was dependent on the duration of incubation, but the binding capacity remained constant after 2 h incubation.
