N2 - Recently we discovered "activation factor" which prevents ATP inhibition of liver phosphofructokinase (Proc. Natl. Acad. Sci., in press). The effect of glucagon and glucose administration on the levels of the factor was investigated using isolated hepatocytes. The concentration of the factor in hepatocytes increased at least 15-fold within 8 min after administration of 20 mM glucose. Upon glucagon administration (10-7M), the level of activation factor decreased rapidly and within 2 min the amount decreased to 1/13 of the original concentration. It is proposed that glucagon acts to inhibit phosphofructokinase in the liver by (1) decreasing the level of activation factor, and (2) phosphorylation of the enzyme.
