A fluid membrane-based soluble ligand-display system for live-cell assays

Abstract

Getting the right exposure. Membrane-tethered epidermal growth factor (EGF) is found to modulate substrate attachment and spreading of human breast epithelial cells by EGF-EGFR signaling interactions. In a newly implemented system, the fluidity of the supported membrane enables localized enrichment of ligand density in a configuration that reflects cognate-receptor distribution on the cell surface. This technique provides a means to control soluble ligand exposure within a parallel-surface array format.
